Core Components

A typical qPCR master mix or reagent kit comprises the following essential components:

  • DNA Polymerase: Thermostable hot-start enzymes (e.g., Taq) that initiate amplification only after the initial denaturation step to prevent non-specific binding.
  • dNTPs: A balanced mixture of dATP, dCTP, dGTP, and dTTP acting as the building blocks for the newly synthesized DNA strands.
  • Reaction Buffer: Optimized buffers containing MgCl₂ and other salts to maintain enzyme stability, pH, and ideal primer-annealing conditions.
  • Detection Chemistries: Either sequence-specific fluorescent probes (like TaqMan) for multiplexing or DNA-binding dyes (like SYBR Green) for general dsDNA quantification.
  • ROX Reference Dye: An internal passive reference dye used in many thermocyclers to normalize well-to-well fluorescent signal variations.
