Typical Workflow

  1. Lysis: Cells are disrupted using detergents and enzymes (e.g., Proteinase K) in the presence of chaotropic salts to release DNA and inactivate nucleases.
  2. Binding: The lysate is passed through a spin column or mixed with magnetic beads; the chaotropic salts and alcohols force the DNA to bind to the silica or bead surface.
  3. Washing: Specialized wash buffers remove cellular debris, salts, and enzymes.1
  4. Elution: Purified DNA is released from the membrane or beads using a low-salt buffer or nuclease-free water, ready for downstream applications.
